Cell-mediated immunogenicity assay research achievements

At the Cytokines & KAI 2024 Conference, JaeWook Hyeon PH.D , Manager of Development Team 2,HyungJung Lee, ELISpot Project Lead, and TaeHyun Kim, Flow Cytometry Project Lead, presented their research findings aimed at enhancing the quality of clinical sample analysis using ELISpot and immune phenotyping analysis of immune cells using PBMC.The first presentation focused on "Optimization of Multiplex ELISpot Analysis Method Using Three Fluorescences." This method evaluates cell-mediated immunogenicity in clinical trial subjects by isolating PBMCs (peripheral blood mononuclear cells) from blood and measuring cytokines secreted by immune cells through ELISpot. By analyzing IFN-γ, TNF-α, and IL-5 simultaneously in a single sample using three distinct fluorescence colors, the method achieved a significant reduction in the required blood volume by two-thirds compared to previous methods.The second presentation was titled "Stability of Immune Phenotyping Analysis in PBMCs After Long-Term Cryopreservation." During clinical trials, PBMCs isolated from blood are sometimes stored and analyzed together to reduce variability between analyses. However, long-term storage can pose a risk of producing variable results depending on the timing of the analysis. Based on practical experience, this study selected representative CD markers and confirmed that PBMCs can be stored at ultra-low temperatures (below -150°C) for up to 24 months without compromising stability.For the inquiry, please send mail to GCCL@gccorp.com.
